Resume
Purpose: to study the methylation features of microRNA genes (miR-107, -124a-1/2/3, -125b-1, -127, -130b, -137, -203a, -375) in different molecular subtypes of breast cancer. Materials and methods. 70 patients are included in research with a verified diagnosis of breast cancer in the age from 30 to 72 years. Patients with stage II (51.4%) and III (31.4%) are prevailed. Metastases to the lymph nodes were observed in 60%. Invasive ductal carcinoma was detected in 72.8% of cases, invasive lobular carcinoma - 12.8% of cases, other forms of cancer were diagnosed in 14.4% of patients. Moderately differentiated breast cancer was in the majority of patients (74.2%), in 21.4% of cases there was low differentiation, 4.4% of patients had a well-differentiated tumor. Luminal subtype A was found in 27.1% of patients; luminal subtype B Her2-negative - in 18.5% of patients; in 32.8% of cases - luminal subtype B Her2-positive, thrice-negative - in 14.2% and non-luminal Her2 (+) positive subtype - in 7.1% of cases. Methyl-specific polymerase chain reaction (MS-PCR) was used to study the methylation profile of microRNA genes (miR-107, -124a-1/2/3, -125b-1, -127, -130b, -137, -203a, -375 ) in paired images of tumor and histologically unchanged breast tissue. The results were processed with the standard packages STATISTICA, v. 10 and IBM SPSS Statistics, v. 21. Results. It was found that in luminal A subtype breast cancer the frequency of miR-124a-1 and miR-125b-1 methylation in tumor tissue increased by 2.3 (84.2% (n = 16) versus 36.8% (n = 7), p = 0.0069) and 8 times (42.1% (n = 8) versus 15.8% (n = 1), p = 0.0189) respectively in comparison with unchanged breast tissue. At Her2-negative and at Her2-positive lyuminalny In a subtype reliable differences in the frequency of methylation of genes of microrNA miR-124a-1, miR-375 in a tumor and histologically are shown to not changed fabric. The methylation rate of these microRNAs at Her2-positive subtype was 69.5% (16/23) versus 17.3% (4/23), (p = 0.0008) and 43.4% (10/23) versus 13% (3/23), (p = 0.0472), respectively. For Her2 - negative cancer, these values were as follows: 84.6% (11/13) versus 38.4% (5/13), (p = 0.01) and 69.2% (9/13) versus 23% (3/13), (p = 0.01), respectively. A high methylation frequency of miR-124a-3 and miR-125b-1 was established at the Her2-positive B subtype of CMW. At the triple-negative subtype, methylation of miR-125b-1 in tumor tissue was observed in most samples (70% (n = 7), p = 0.0075), and was absent in histologically unchanged tissue. Conclusion: All studied miRs demonstrated increased levels of methylation of the CpG islands of the promoter regions which indicates their potential diagnostic value. The frequency of miR-375 methylation was significantly increased in tumors in the triple-negative subtype in comparison with the luminal B Her2-negative BC subtype. For the Her2-positive luminal B subtype was noted a high percentage of 6 miR methylation.
