AGE-RELATED CHARACTERISTICS OF MAST CELLS IN IMMEDIATE-TYPE ALLERGIC REACTIONS

Aim of the study: To identify age-related characteristics of mast cell reactivity in allergies and develop criteria for therapeutic efficacy based on an experimental model of specific hyposensitizing immunotherapy. Materials and Methods. A total of 443 guinea pigs of both sexes and ages were used, after undergoing a 10-14-day quarantine period in a vivarium. The animals were constantly monitored, with periodic body weight and rectal temperature measurements. Sensitization and anaphylactic reactions were simulated using standard methods. Normal horse serum was used as the allergen. A single challenge dose of the specific allergen was administered intravenously or intraperitoneally 14-16 days after the initial sensitizing dose of the allergen. The collected digital data were processed using Microsoft Excel. Results. The rate of mast cell granule filling increases with age. In older guinea pigs, compared with newborns, the rate of mast cell granule filling was 1.3-2.2 times higher (p < 0.05), which is consistent with published data. Spontaneous mast cell degranulation was observed in intact animals. Their frequency ranged from 5.4 to 8.6% and was essentially independent of age. A comparative analysis of degranulation frequency depending on age revealed that the degranulation frequency in sensitized neonates was somewhat lower than in older animals. However, with age, the frequency of degranulated mast cells increased in sensitized guinea pigs, and mast cell reactivity in month-old guinea pigs became approximately the same as in mature animals.
