EFFECT OF MINIGASTROBYPASS ON METABOLIC PARAMETERS IN RATS WITH ALIMENTARY OBESITY AND ALLOXAN-INDUCED DIABETES

Resume
Objective – to comparatively assess the dynamics of body weight, carbohydrate and lipid metabolism in rats with diet-induced obesity and alloxan-induced diabetes mellitus subjected to OAGB versus non-operated animals. Materials and Methods. Fifty-seven male Wistar rats were allocated into three groups: intact animals (n=12); rats with diet-induced obesity and alloxan diabetes without surgical intervention (n=22); rats with the same combined pathology subjected to MGB with a 25-cm biliopancreatic limb (n=23). Observation time-points were days 7, 14, and 30. Body weight, fasting glycemia, glucose tolerance with calculation of the area under the glycemic curve, and lipid profile (triglycerides, total cholesterol, HDL-C, LDL-C) were assessed. Results. By day 30 body weight in the MGB group decreased by 11.3±1.4%, whereas non-operated diabetic animals showed a modest decrease of 3.1±0.9% (p<0.001). Fasting glycemia in the operated group fell from 13.5±0.6 to 6.2±0.4 mmol/L (p<0.001), while remaining persistently high (13.6±0.5 mmol/L) in non-operated animals throughout the study. The area under the glycemic curve during OGTT was 1180±95 mmol/L·min in the MGB group versus 2640±180 in non-operated diabetic rats (p<0.001). Triglyceride and total cholesterol levels in the MGB group decreased by 43.8±3.9% and 38.1±3.6%, respectively (p<0.001). Conclusion. Mini-gastric bypass in a model of diet-induced obesity and alloxan diabetes is associated with significant body weight reduction, normalization of fasting glycemia, restoration of glucose tolerance and improvement of the lipid profile, supporting the pronounced metabolic effect of the procedure.
