Objective: To compare the number and concentration of platelets, the number of residual erythrocytes and leukocytes in platelet-rich plasma samples prepared by single and double centrifugation methods. Material and methods: 80 healthy people (30-60 years old; 40 women, 40 men) were divided into two observation groups of 40 people each. 20 ml of venous blood was collected from all those observed. In group 1, to obtain platelet-rich plasma, a single centrifugation of blood samples was performed, and in group 2, a double centrifugation of blood samples was performed. The composition of platelet-rich plasma obtained by these methods was compared. Results: the average platelet count in group 1 was 589.3±285.1×103/μl, and in group II – 918.11±289.79×103/μl. The average concentration/yield of PRP platelets in group 1 was 168.69-54.18%, in group II - 255.19-221.6%. Differences between groups in platelet count, platelet concentration/yield, and residual white blood cell count were statistically significant (p<0.01), but differences in residual red blood cell count were not significant. Conclusion: the double centrifugation method turned out to be more effective compared to the single centrifugation method, since it made it possible to obtain a significantly higher number and yield of platelets in platelet-rich plasma samples, as well as a significantly lower number of residual leukocytes.
